Estimates suggest that greater than 85% of the human genome is “undruggable” using traditional small molecule therapies. mRNA medicines promise to overcome this limitation, but only if they can be delivered to the correct location in the body, at the right dose, and for the appropriate amount of time. Here, we provide an overview on strategies and materials that can be used to improve the delivery of mRNA in vitro and in vivo, with the ultimate goal of providing fundamental background information and translational information on the use of mRNA based drug candidates.
